Reported mechanism

Agonist of all melanocortin receptors (MC1R through MC5R) with highest activity at MC1R in melanocytes. MC1R activation drives cyclic AMP signaling, tyrosinase upregulation, and increased eumelanin synthesis. The D-Phe⁷ substitution is the key modification that increases potency and protease resistance.

Regulatory status

FDA-approved as Scenesse (afamelanotide) for erythropoietic protoporphyria (EPP) — a rare inherited condition causing painful photosensitivity. The implant formulation is tightly restricted. Research-grade material is for non-clinical use only.
